Practical Training of the RµI Network

Objectives

  • Discover and handle various tools and techniques for preparing frozen biological samples and Cryo-microscopy.
  • Prepare and observe biological (plant) samples under the electron beam of a FEG-SEM equipped with Cryo-microscopy or with a cooled stage (observations in controlled pressure mode with a Peltier-effect stage).
     

Target Audience

Priority is given to members of the INRAE Microscopists Network (RµI) and INRAE staff involved in Cryo-SEM imaging.
Prerequisites: Engineers, technicians, or researchers with good experience or knowledge of scanning electron microscopy (SEM) tools.

Program

  • Theoretical Input:
    Freezing of water and hydrated samples
    Techniques and equipment in Cryo-microscopy
    Handling of liquid nitrogen and cold objects
  • Practical Workshops:
    Observation in controlled pressure mode with cooled stage
    Cryo-sample preparation and observations under SEM electron beam
    Rapid freezing, fracture, vacuum sublimation, cold metallization
    High-resolution observations, micro-analyses (if possible)
     

Equipment

  • Peltier-effect cooled stage (-50°C / +50°C)
  • ACE 600 Cryo-coater
  • Cryo-stage + VCT 100 cryo-transfer module
  • Sigma HD VP FEG-SEM + EDS spectrometer
